Abstract

The main goal of this thesis was the analysis of new methods for electron microscopy grid preparation and protein isolation focused on reducing the required sample volume without sacrificing EM grid quality and resolution, and on the application of cryo-EM and 3D reconstruction procedures for various single particle projects, including native human 20S proteasome, tobacco mosaic virus, interphotoreceptor retinoid-binding protein, and leucine-rich repeat kinase 2.
