Abstract
This thesis was focused on the development and application of new methods using LC-MS/MS for the quantification of antibiotics, with a special focus on beta-lactams. Different approaches in HPLC, including HILIC, reversed phase and 2D column systems, were used. The applicability of the developed methods for Therapeutic Drug Measurement in plasma and filtered plasma as well as measurements in different tissues was shown. The influence of filtering techniques on the resulting free fractions from filtered plasma was investigated and recommendations could be made for future clinical use. The methods were also used in stability indicating studies for different pharmaceutical dosage forms as well as in studies on the clinical efficacy of beta lactam measuring.
