Abstract

The first part of the introduction presents artificial cofactors that are employed for the formation of streptavidin-based artificial metalloenzymes. Their interaction with the protein scaffold is illustrated and compared. This part is intended as a first orientation on Sav-based artificial metalloenzymes and contains a summary of common features and considerations for the development of versatile cofactors. Screening strategies are briefly outlined, and the relevant literature is referenced for the interested reader. The second part of the introduction focusses on the actual streptavidin constructs that are used in the Ward group and is followed by highly detailed materials, methods, and notes sections for the large-scale expression of Sav-variants.
